چکیده

The proteinase-catalysed synthesis of [Leu]enkephalin and [Met]enkephalin was studied kinetically. Na-t-Butoxycarbonyl-amino acids and peptides or their ethyl esters served as acyl donors, and amino acid phenylhydrazides were used as acyl acceptors. Initial-velocity measurements of oa-chymotrypsin-catalysed peptide synthesis gave rise to kinetic patterns that are compatible with a ping-pong mechanism modified by a hydrolytic branch. Initial-rate and alternative-substrate inhibition patterns for papain-controlled peptide-bond formation are consistent with a sequential ordered mechanism with the acyl donor as the obligatory first substrate. On the basis of the observed kinetic features, reaction mechanisms are proposed for chymotrypsinand papain-catalysed peptide synthesis that inversely equal those describing the pathways of proteolysis. The respective initial-velocity expressions for bireactant systems are given, along with the numerical values of the corresponding kinetic parameters.

Proteases as catalysts for enzymic syntheses of opioid peptides.

Two alternative pathways leading to protease-mediated syntheses of Leu- and Met-enkephalin are described. Each peptide bond of the opiate peptides was formed by either papain, or alpha-chymotrypsin catalysis. N alpha-t-Butyloxycarbonyl amino acids and peptides or their esters served as carboxyl components, whereas amino acid and peptide phenylhydrazides were used as acceptor nucleophiles. After...

Acid Catalysed Condesation of Phloroglucinol and Acetone-Kinetics and Mechanism

The kinetic study of the condensation reaction between phloroglucinol and acetone was investigated at 30, 35, 40 and 45±0.05 °C in 100% methanol. The hydrochloric acid concentrations used were 0.0261, 0.0364, 0.0577 and 0.0728 M. The reaction was investigated with and without a promoter, thioglycollic acid (TGA), and taking into account the functionality (h<e...

Triphenylphosphine Catalysed Facile Multicomponent Synthesis of 2-Amino-3-Cyano-6-Methyl-4-Aryl-4H-Pyrans

Triphenyl phosphine(PPh3), an efficient and reusable catalyst, catalysed the synthesis of 2-amino-3-cyano-6-methyl-4-aryl-4H-pyran-5-ethylcarboxylate derivatives by one-pot condensation of aromatic aldehydes, malononitrile and ethyl acetoacetate in EtOH-H2O (1:1) at reflux conditions. The results show that ...

Efficient Synthesis of Biodiesel From Waste Cooking Oil Catalysed by Al2O3 Impregnated with NaOH

Due to the high price of virgin vegetable oils and the drawbacks of the homogeneous catalytic transesterification, in this work an economically profitable alternative process was proposed for biodiesel synthesis in which transesterification of the low-cost waste cooking oil (WCO) with methanol in a heterogeneous system was done. Alumina impregnated with sodium hydroxide was utilized as a solid ...

Chemical synthesis and papain-catalysed hydrolysis of N-alpha-benzyloxycarbonyl-L-lysine p-nitroanilide.

The chemical synthesis of N-alpha-benzyloxycarbonyl-L-lysine p-nitroanilide (Z-Lys-pNA) is described in detail. The pH-dependence of the catalytic parameters kcat,' Km and kcat./Km for the papain-catalysed hydrolysis of Z-Lys-pNA are determined. kcat. and Km are pH-independent between pH 5 and pH 7.42, but the pH-dependence of kcat./Km is bell-shaped, decreasing at high and low pH values with p...
